HZAU Makes New Progress in 3D Genome Architecture

On November 14, Professor Li Xingwang and Professor Li Guoliang of the National Key Laboratory of Crop Genetic Improvement of HZAU published a review article entitled “Technologies for Capturing 3D Genome Architecture in Plants” online in Trends in Plant Science.

3D genomics is one of the hotspots and frontiers of genomics. At present, biologists have developed a lot of high-throughput 3D genomics research technologies for human cell lines and animal cells, some of which have been successfully applied to plants.

3D genomics research technologies

The team of Prof. Li Xingwang and Prof. Li Guoliang has been committed to the research of 3D genomics and epigenetics. In the early stage, they developed two technical systems (long-read Chia pet and DLO Hi-C) with human cell lines and animal cells as research materials. In 2019, they established the Long-read ChIA-PET technology system in maize and rice for the first time, and depicted the 3D genome map of them. In 2020, they used DLO Hi-C technology to illustrate the 3D genome of male and female ears of maize as well.

This review paper introduces the research techniques of plant 3D genomics. It first briefs the technologies of 3C, Hi-C, in situ Hi-C, DLO Hi-C, ChIA-PET and Hi-ChIP applied to plant 3D genomics research, and then elaborates on how to use these technologies to analyze the 3D structure of plant genome with special attention to the chromatin loop structure detected by ChIA-PET and Hi-ChIP. The advantages and challenges of current plant 3D genomics research technology are also proposed, which will have great significance for the future development of plant 3D genomics technology and the analysis of 3D genome structure.
